20회 microTAS 학회의 시작을 알리고자 ‘opening remarks’를 준비 중이다.
약 1,000석 가까이 되는 좌석이 마련되어 있다.
I. 주요 발표 내용1. 10월 10일 주요 내용· Plenary I.Precision ‘reading’ and fabrication of surface architecture on nanoscale (Kenneth A. Dawson, University college Dublin, Ireland)
1) Synthetic nanoparticles: 생물학적 환경에서 발생하는 synthetic nanoparticles는 자연적으로 발생하는 biomolecular assemblies 만큼이나 origin의 생물학적 상태나 기능을 이해하는데 도움을 준다. Exosome과 같은 extracellular vesicles가 대표적이며, diseases를 진단하는데 이용할 수 있는 새로운 liquid biopsy 물질로서 주목을 받고 있다. 하지만 이를 detection 하는데 많은 한계를 지니고 있다.
2) Endogenous processes are nanoscale; Engineered nanoscale written in our biology: 저녁을 먹거나 감기에 걸리거나 암 발생 등의 모든 것은 nanoparticles에 의해 반영되고 특히 대사작용 담당하는 liver의 경우 약 40% 정도의 nanoparticles가 축적된다. 각각의 surface에는 특징적인 biomolecules를 발현하며 따라서 모든 particle은 ‘reading’ 가능하다 (Make and measure in small spaces, make and measure ‘one by one’).
3) The development of platforms for screening the exposed epitopes of biomolecules on the surface of nanoparticles: Nanoparticle을 포획하기 위해 표면에 특정 epitope과 반응하는 antibody를 이용하는 방법이 주로 사용된다. Antibody에는 gold나 quantum dots와 같은 small-particles를 conjugation하여 nanoparticles의 visualization, quantification, spatial localization을 가능케 한다. 특히 lab-on-a-chip system을 적용한 microfluidic flow 기반 기술은 small detection volume으로도 nanoparticles의 distribution 및 quantification을 가능하게 하였으며 실험적 정밀성(precision)에서의 향상을 가져왔다.
· Session 1.Simple and rapid formation of 3D co-culture cell laden microstructures by using cell origami technique (Q. He, Hokkaido university, Japan)
1) Three dimension (3D) co-culture:
In vivo 상태에서는 여러 종류의 cells가 interaction을 하면서 cell functions와 proliferation을 담당하고 있으며, 이를
In vitro 상에서 cell을 위한 microenvironments을 제공하기 위해서는 monoculture가 아닌 3D co-culture system을 적용할 필요성이 있다. 기존의 3D co-culture 방법들 (microfluidic + shaking, microfluidic device, magnetic force methods 등)은 복잡하여 시간이 많이 소모되는 단점을 가지고 있다.
2) Cell origami method: Microplate 기반의 3D cell co-culture microstructure를 제작하여 origami(종이접기) 방식으로 cell을 배양하는 기술은 기존 대비 빠르고 간단하다는 장점을 가지고 있다. 3단계로 구분되는데, 다음과 같다.
① Making microplates (3D로 접을 수 있는 design을 선택함)
② Seeding cells (첫 번째 cells를 seeding 후 washing 과정을 거쳐 두 번째 cells를 seeding 함)
③ Folding (구형으로 folding을 하면, 첫 번째 cells는 외부에, 두 번째 cells는 내부에 위치함)
특히, “sacrificial layer”을 만들어 한번에 seeding이 가능한 3D structure의 수량을 증가시켰고, 시간 control을 가능하게 하였으며 folding이 신속하게 일어나도록 했다.
3) Cells in origami structure: Folding 후 3D microstructure 내부를 날마다 관찰한 결과, 6일까지 cells가 생존하는 것으로 확인되었다. 기능적인 측면을 확인하기 위하여 folding 7일 후 co-culture cells의 albumin secretion을 unfolding co-culture system과 비교하였을 때, 유의적으로 1.5배 이상의 secretion 량이 증가했음을 확인하였다. 이후에는 다양한 모양의 microstructure의 개발 및 co-culture cells와 microplate의 분리율 향상, 3D co-culture structures의 size 확대가 향후 개발 목표이다.

2. 10월 11일 주요 내용· Plenary III.Nanostructures for sensing - Applications in health (Anja Boisen, Technical university of Denmark, Denmark)
1) Nanostructures for sensing: Silicon으로 만든 nanopillar substrates, nanomechanical resonators, nano-sized magnetic beads, microfabricated electrodes 등을 이용한 여러 종류의 sensor이 개발 가능하다. 각각을 이용함에 있어서의 장단점이 존재하고 한 sample에서 동시에 size가 다른 물질을 분리하는데 적용하기 위해서는 방법적 combination이 필수적이다. Human health 분야에 sensors를 개발하였고, 이는 patient monitoring, diagnostics, material characterization 등에 적용 가능하다.
2) Cantilever based sensing: Micrometer sized cantilever는 한쪽에 probe molecule을 가지며, 이곳에 target이 붙음에 따라 surface stress의 변화에 의해 cantilever는 굴절이 되는 원리를 이용한다. Cantilevers 수백 개를 rotating disc system으로 만들어 target binding에 따른 굴절을 1초 안에 읽을 수 있도록 만들었다. 본 sensor는 biomarker를 detection하는데 사용할 수 있다.
3) Vibrating micrometer sized strings: 효율적이고 민감하게 mass detection이 가능하여 single nanoparticle의 chemical 분석에 이용할 수 있다. Micrometer sized strings의 resonance frequency shift나 polymer로 얇게 coating된 cantilever를 monitoring 함으로서 nanogram 정도의 변화도 알아낼 수 있다. 본 sensor는 biomaterials의 degradation 확인이나 나아가 미세한 온도 변화 등에도 적용될 수 있다.
4) Nanopillar SERS (surface enhanced Raman spectroscopy) substrates: Glass와 silicon으로 개발하였고 Raman signal을 높고 uniform으로 향상시킬 수 있다. 간단한 방식의 fabrication은 SERS를 기반으로 한 sensing에 있어 재현성 있는 platform을 제공한다는 측면에서 의미가 있다. 본 sensor는 cell supernatants나 patient breath와 같은 small analyte detection에 사용할 수 있다.
· Session 2.Comprehensive molecular profiling of single circulating tumor cells from lung cancer patients (Seung-min Park, Stanford university school of medicine, USA)
1) CTCs (Circulating tumor cells): Genomic, transcriptomic, proteomic, cellular components 등을 포함하는 blood-based biomarker는 cancer detection 및 assessment에 있어 중요하다. CTC는 tumor로부터 release되어 bloodstream으로 흐르는 tumor cells을 의미하며, 기존 biopsy 방법이 아닌 liquid biopsy 방법으로 cancer management를 할 수 있다는 측면에서 활발히 연구되고 있다. 2004년 EpCAM antibody를 이용해 CTC를 분리할 수 있는 장비인 “CellSearch”가 FDA 허가를 받았지만, 이 장비는 완전한 모양의 CTC가 아닌 irregular CTC나 apoptotic CTC 등은 확인이 어려운 단점을 가지며, 이는 CTC 염색의 기본 원리인 immunocytochemistry (ICC) staining의 한계이기도 하다. 또한 CTC counting만 가능하고 이후 CTC analysis가 불가능하여 암환자 치료에 응용되기 힘들다.
2) MagSifter (Magnetic sifter): EpCAM antibody를 통한 분리를 목적으로 제작하였다. 12 μm NiFe film으로, 40 μm 직경의 hole이 존재하는 구조로 standard lithography, high magnetic field gradient의 원리를 적용하였고 10 mL/h라는 high efficiency cell capture가 가능하다.
3) Nanowell: Cell 분리 후 CTC 확인을 위해 제작되었으며 25,600 wells로 구성되어 있고 Si wafer에 PDMS를 적용한 구조이다. 많은 wells에 의해 single cell seeding이 가능하며 RT-PCR reagent를 넣은 후 증폭까지 가능한 lab-on-a-chip이다.
4) Nanowell validation: NSCLC 및 non-cancer cell line으로 진행하여 nanowell의 성능을 validation 하였으며, ICC 방법과의 비교를 통해 sensitivity가 향상되었음을 보여주었다. 20명의 healthy blood와 23명의 advanced stage NSCLC blood로 CTC 분리를 진행했을 때 cutoff는 7개로 나타났으며, 4명의 sample로 RT-qPCR를 진행하여 EGFR mutation을 확인했을 때 biopsy 결과와 일치함을 확인하였다.
3. 10월 12일 주요 내용· Plenary V.Can engineered micro-scale organotypic models predict patient-specific responses? (David J. Beebe, University of Wisconsin, USA)
1) Micro-scale organotypic models: Micro-scale organ-on-a-chip model을 통한 patient-specific cancer response를 확인하고자 하는 노력들이 있다. 이를 이용하여 ① Patient로부터 얻어진 multiple myeloma cells와 stroma를 가지고 co-culture를 진행하여 약물의 chemosensitivity를 예상하며, ② ECM으로부터 prostate cancer cells가 invasion하는 것을 이용한 환자의 분류를 진행하고, ③ 환자의 endothelial cells를 이용하여 organotypic vessels를 만들어 anti-angiogenic therapy의 감수성을 확인한다.
2) In vitro to ex vivo-Blurring the lines: In vitro model (glass 위에서 일어나는 events)과 ex vivo samples (clinical 연구와의 융합)를 이용하여 multi-omic & functional orthogonal endponts를 도출하고, 환자의 monitoring을 통한 treatment decision을 가능케 하며, 환자의 outcome은 endpoint의 cancer cell 개수로 예상 가능하다. 예상 model을 만들 때, 중요한 points는 ① Model로부터 얻어진 결과에서 포함시킬 것과 뺄 것은 무엇인지 결정해야 하며, ② 어떠한 방법으로 model을 validation 할 것인가 고민해야 하고, ③ 최적의 endpoint를 무엇으로 정의할 것인지를 정해야 한다는 것이다.
3) Open-Lego type: 보통의 microfluidic system을 적용한 lap-on-a-chip의 고민 points는 bubble 발생시 제거를 어떻게 할 것인가와 하판과 상판의 sealing을 어떻게 하여 chip의 내구성을 유지시킬까 하는 것이다. Lego 방식을 응용한 open type의 microfluidic chip은 기존의 문제점을 극복한 것으로, 이를 이용하여 적은 환자 sample로부터 cell, DNA, mRNA, protein을 분리하여 downstream 연구가 가능하다.
· Session 3.A digital microfluidic platform for non-invasive prenatal genetic diagnostic screening(M. D. Chamberlain, University of Toronto, Canada)
1) Digital microfluidic (DMF) platform: Channel이 아닌 electric fields를 이용하여 actuation electrode와 ground electrode 사이에 있는 각각의 droplets를 조절(dispense, move, merge, mix, split 등) 가능하도록 만든 platform이다. Hydrophilic surface를 적용하여 cell이 attachment되어 growth가 가능하도록 설계하였다. Laser microbeam cell lysis (LCL) 기술을 적용하여, single cell 이나 원하는 cell을 lysis하여 downstream 분석을 가능케 하였다.
2) Prenatal genetic test: 5,000개 이상의 genetic mutation이 존재하며 1/33의 비율로 genetic mutation이 발생한다. 이를 진단하기 위해 amniocentesis가 행해지고 있지만, highly invasive한 방법으로는 mother 뿐만 아니라 fetus에도 위험하다. 이를 극복하고자 non-invasive한 방법으로 cell-free fetal DNA (cffDNA)을 분리하여 이용하고 있지만, Fragmented DNA로 부분적인 abnormalities (duplication of a chromosome)만을 알 수 있고 Trisomy 13, 18, 21과 sex aneuploidy 와 같은 major abnormalities만을 구별한다는 한계점을 가지고 있다.
3) PAP-smear collection: 의사가 sample 수집이 편하고 minimally invasive하며 mother와 fetus에게 위험이 없다는 장점을 가진다. Fetal cell type 중 trophoblast를 targeting 했으며, 초기 test와 모든 genetic mutation을 목표로 연구를 진행하였다. 분리된 trophoblast는 HLA-G, cytockeratin 7, CD227로 staining하여 구별하였다. 이후 DMF-LCL을 이용하여 fetal cell의 lysis를 진행하여 수 nL의 droplet으로 collection 한 후, whole genome amplification을 통해 electrophoresis 및 sequencing을 진행하여 genomic abnormalities를 확인하였다.
4. 10월 13일 주요 내용· Session 4.Micro-well array based genetic determination of EGFR mutation at single cell level (Ren Li, National center for nanoscience and technology, China)
1) EGFR mutation in lung cancer: Lung cancer는 매년 14%의 new case가 발생하며 28%는 lung cancer로 죽는다. Non-small cell lung cancer (NSCLC)의 경우, EGFR을 표적으로 하는 drug가 많이 개발되어 있기에 이를 처리하기 위해서는 EGFR의 mutation (Exon 18&20, 19, 21)을 확인해 볼 필요가 있다. Drug sensitive는 L858R과 E749-A790 deletion이 확인되고, drug resistant는 T790M이 확인된다.
2) Detect gene mutation and single cell level: Single cell로 분석을 진행해야 하는 이유는 다음과 같다.
① 각각의 cells는 heterogeneous하여 alternate하기 때문이다.
② 집단으로 분석을 할 경우 information이 covered될 가능성이 있다.
③ 분석의 accurate와 fast를 위해 필요하다.
본 연구는 single cell tapping, cell identification, cell selection & collection, gene amplification, gene sequencing의 순서로 진행되었다.
3) Double-sided micro-well array microfluidic chip: Microfluidic channel을 유체가 통과하면서 아래에 있는 micro-wells에 single cell이 위치하도록 chip이 설계되었으며, CCD를 이용해 imaging 및 identification이 가능하다. Lysis reagent를 넣어 cell lysate collecting chamber에 lysate가 collection 되도록 했고, 이를 회수하여 amplifying 및 sequencing을 진행한다. EGFR mutation을 가지고 있는 cell line으로 test를 해 보았을 때 single cell 단위에서 mutation을 확인하였다.

II. 총평올해로 20회를 맞이한 MicroTAS 학회는 그 동안의 업적을 소개하는 공간을 배치(history exhibition)하여 역사와 함께 진행되었다. 특히 공학 중심의 학회에서 벗어나, bio와의 융합을 시도한 연구가 눈에 띄게 증가한 것이 특징이다. 약 1,200명이 참석했으며 일본, 미국에 이어 한국이 3번째(100명)로 많은 참석국가로 다수의 연자 발표 및 포스터 발표가 있었다. 마지막 날에는 microfluidics 분야에서의 저명한 학술지인 Analytical Chemistry와 Lab on a Chip에서 award 진행 및 poster와 video award를 진행하는 등 연구자들을 응원했으며, 내년 미국 사바나에서 만날 것을 약속하면서 4일간의 학회는 막을 내렸다.
아일랜드 독립 100년을 기념하는 건축물이 곳곳에 보인다.
리피강을 중심으로 명소들이 위치해 있어, 관광지로도 각광을 받고 있다.
